Paris Centre de Recherche Cardiovasculaire (U970)
The PARCC (Paris Cardiovascular Research Center, Inserm U970) brings together a critical mass of basic science researchers and clinicians who share a common vision and common objectives in the field of cardiovascular diseases. The PARCC is part of Université Paris Cité and its Institute of Cardiovascular Sciences. Team Bouatia-Naji is member of the iWiSH (interdisciplinary institute of women’s health).
Team Bouatia-Naji gathers clinicians, statistical and molecular geneticists, and cell biologist to improve knowledge about cardiovascular disease predominant in women. We study the genetic causes and their biological translation in fibromuscular dysplasia (FMD) which manifest typically in early to middle adulthood in women in the context of stroke, hypertension of myocardial
infarction due to SCAD. FMD is an abnormal narrowing or bulging (stenosis and aneurysms) in medium-sized arteries. Unlike atherosclerosis, FMD is not caused by plaque build-up or inflammation, and its biological mechanisms remain largely unknown. Progress has been accelerating since our genetic discoveries related to smooth muscle cell contraction function and the role of extracellular matrix biology. Modelling of recently discovered common and rare genetic variation involved in FMD risk will be key to fully understand its specific biology, and set the
stage for targeted therapeutics.
The postdoc position is part of a Leducq Network of Excellence (Align-FMD, see website). Partner Bouatia-Naji is leading the induced pluripotent stem cell-derived SMCs differentiation of genetic
variation associated with FMD, in loss-of-function, gain-of-function and variant-specific models.
Effects on SMCs and their ECM will be evaluated through functional endpoints, including migration, proliferation, adhesion, and ECM synthesis. Testing will be conducted in variant specific and gene specific models, utilizing genetic methods to alter candidate genes’ DNA sequence (CRISPR/Cas9) and/or expression levels, after nomination from the human genetic studies that is an important part of this project. The postdoc is expected to involved in this specific activity of this Leducq project, participate in an international context of research in interaction with members of the network.
Main activities
The postdoctoral researcher will work with iPS cell lines and their differentiated smooth muscle and endothelial cells, if applicable, ensuring their maintenance and daily follow-up.
The position also involves designing and refining cell-transfection approaches, exploring different methods to identify the most effective strategies for the team’s experiments. The researcher will routinely use core molecular biology techniques to support mechanistic studies related to vascular function. A central part of the work will also consist in characterizing cell populations through immunostaining and advanced imaging approaches, using optical, fluorescence, and confocal microscopy, as well as flow cytometry. Finally, the postdoctoral researcher will engage with supporting members of the lab involved in molecular and cell biology, including students, communicate about progress and propose analytical and research experiments. Leducq networks require high
level of communication skills in a large collaborative work environment.
Profil recherché
Desired Experience
• 3 to 5 years (PhD + postdoc, eventually) in vascular biology, including experience with iPSC
genetic modification and cell culture, and a strong background in molecular biology.
Degree Level and Education
• PhD in vascular biology or cell biology in the context of human disease.
